Transgenic mangium trees (Acacia mangium) overexpressing xyloglucanase (AaXEG2) were generated by spraying flower buds with Agrobacterium solution and allowing seeds to develop. The overexpression of xyloglucanase decreased xyloglucan content in the cell walls and increased stem length and diameter. The leaves of the transgenic seedlings exhibited accelerated heteroblastic development, proceeding from the stage of three bipinnate leaves to that of enlarging petiole 2 weeks earlier than wild type seedlings did.  相似文献

We investigated the impact of land-use changes on the soil biomass at several soil sites in Indonesia under different types of land-use (primary forest, secondary forest, coffee plantation, traditional orchard, and deforested area), located within a small geographical area with similar parent material and climatic conditions. Various parameters of soil microbial biomass (biomass C, biomass N, content of anthrone-reactive carbohydrate carbon, and soil ergosterol content) were examined. Our results suggested that the removal of the natural plant cover did not cause any appreciable decrease in the amount of microbial biomass; on the contrary it led to a short-time increase in the amount of microbial biomass which may be due to the availability of readily decomposable dead roots and higher sensitivity to the decomposition of residual litter in recently deforested soils. However, the amount of microbial biomass tended to decrease in proportion to the duration of the land history in coffee plantation soils. This may be ascribed to the effect of the loss of available substrates associated with soil erosion in the long term. Lower ergosterol contents in recently deforested areas reflected a reduction in the amount of fungal biomass which may be due to the destruction of the hyphal network by the slash and burn practice. On the other hand, the higher soil ergosterol content at the sites under bush regrowth indicated that microbial biomass was able to recover rapidly with the occurrence of a new plant cover.  相似文献

Field experiments were conducted at the Indian Agricultural Research Institute, New Delhi, India for three years from 2001–2002 to 2003–2004 to study the relative efficiency of diammonium phosphate (DAP) and mussoorie rock phosphate (MRP) in a rice-potato-mungbean cropping system. Phosphorus application significantly increased productivity, protein yield and energy output of rice-potato-mungbean cropping system and resulted in an increase in 0.5 M sodium bicarbonate (NaHCO3) extractable phosphorus (P) content in soil. The MRP at 35 kg P ha?1 was at par with 17.5 kg P ha?1 as DAP in terms of productivity, protein yield, and energy output but significantly superior in terms of PSB population in soil. Phosphorus balance (application – crop removal) was generally more positive for MRP than DAP and the highest with an application of 52.5 kg P ha?1 as MRP. Present study indicates that P requirement of a rice-potato-mungbean cropping system can be met with 76–79% higher dose of MRP as compared to DAP.  相似文献

Boron deficiency is a major production constraint of Mango cv Alphonso in western coast, Maharashtra, India. The soils are sandy-loam, light, acidic in reaction and receive high annual precipitation of above 2,500 mm. The leaf and soil B status was low with an average of 23 and 0.25 mg kg−1 respectively. About 60% of the orchards were found to be deficient in B. The B adsorption study using Langmuir adsorption isotherm revealed that the adsorption capacity of B by these soils was low with an average adsorption maxima (b) of 16.62 μg g−1 and bonding energy constant ( K ) of 0.09 mL μg−1 making soil B susceptible to leaching. The Mango leaf N status was in excess range, which aggravated B deficiency through growth dilution. A significant negative correlation was observed between leaf B and leaf N (statistically significant at 1% level (p=0.01)). The free Al status of the soils was found to be fairly high, and its detrimental effect on root growth was observed which led to inefficient absorption of limited soil B by roots. The climatic conditions of Konkan like high humidity, high temperature and long sunshine hours also favored widespread incidence of B deficiency, low yield and poor quality. The response of Mango cv Alphonso to the B application was found to be more in foliar than in soil application.  相似文献

Traditional AOAC colorimetric procedures for carotenoid analysis are known to lack specificity and accuracy. Newer HPLC methods provide the investigator with a more precise tool for carotenoid quantification in foods and tissues. In the present studies, reverse phase HPLC was utilized to evaluate the - and -carotene content in raw and cooked leaves of lettuce, spinach and winged bean as well as in the carrot root. The vegetables were boiled or steamed and the true retention of - and -carotene in the cooked products was determined. Boiling for 30 minutes resulted in a 53 and 40% loss of -carotene from lettuce and carrots, respectively. Full retention or even an increase in -carotene content in boiled winged bean leaves and spinach was noted. Steaming resulted in very good retention of - and - carotene in all vegetables (83–139% retention). Thus, although cooking procedures (especially boiling) may result in oxidative loss of carotenoids in some vegetables, heat treatment increases the chemical extractability of - and -carotene in others. The presence of carotenoproteins in some vegetables may effect the heat stability of extractability of - and - carotene.  相似文献

Toxoplasma gondii is a persistent protozoan parasite capable of infecting almost any warm-blooded vertebrates. SAG1 (p30) is the prototypic member of a superfamily of surface antigens called SRS (SAG1-related sequence). It constitutes the most abundant and predominant antigen. In this paper the primary structure of mature SAG1 gene of an Indonesian T. gondii isolate is described and sequence comparison is made with published sequence data of 7 other strains or isolates. Sequence comparison indicated that SAG1 is highly conserved through evolution and despite parasite spreading world-wide. Sequences may be divided into two major families, independent of the strain/isolate geographic origin. Variations were mainly localized at the C-terminal half or domain 2 and some clustered in restricted areas. Sequence comparison allowed us to define the Indonesian isolate as genuine virulent RH strain. A phylogenetic tree of Toxoplasma strains/isolates was constructed based on SAG1.  相似文献

The aim of this research was to identify the changes in the cytoplasmic ultrastructure of immature and matured oocytes in buffalo (Bubalus bubalis ). Oocytes were matured in vitro in tissue culture medium?199 with and without sericin, and then analyzed by light and transmission electron microscopy. The experiment result showed that the nuclear maturation rate of buffalo oocytes was significantly higher in the presence of sericin (80.6%) than without sericin (68.1%) (P  < 0.05). The immature oocytes were characterized by cortical granule clusters in the ooplasm and the absence of perivitelline space (PVS ). In contrast, the oocytes matured either with or without sericin showed the formation of PVS , erected microvilli, the migration of cortical granules to the cytoplasmic periphery, and the clear appearance of the mitochondria and vesicle in the oolemma. Interestingly, matured oocytes with sericin have smaller cortical granules than do immature oocytes (P  < 0.05). In conclusion, supplementation of 0.05% sericin in the maturation medium can enhance the maturation rate of buffalo oocytes. Several cytoplasmic ultrastructures were relocated and modulated during the in vitro maturation process of buffalo oocytes: PVS development, cortical granules migration to periphery, and mitochondria and vesicles in the cortical region. The ultrastructure was similar between the groups with and without sericin.  相似文献

Coelomycetous fungi were screened for the production of TAXOL. TAXOL production of Pestalotiopsis breviseta fungi is confirmed by Ultra Violet (UV) spectroscopic analysis, Infra Red (IR) analysis, high performance liquid chromatography (HPLC), nuclear magnetic resonance (NMR) and LC–MASS spectroscopy. TAXOL isolated from the P. breviseta fungus was identical with authentic TAXOL and produces 0.064mg/L (0.128% dry weight of fungal mat).  相似文献

Growth characteristics [stem diameter (D), tree height (H), and bole volume (V)], stress-wave velocity (SWV), and Pilodyn penetration (P) were measured for 21 seed provenances of 24-year-old teak trees planted in Indonesia to characterize variation in tree growth characteristics, SWV, and P among seed provenances. Broad-sense heritability and correlations between the measured characteristics were also determined. Significant differences for all measured characteristics were observed among provenances, indicating that these characteristics are genetically controlled. Broad-sense heritabilities of growth characteristics, SWV, and P are moderate values. These results indicate potential for improving growth characteristics and wood properties of teak trees with the help of breeding programs. Highly significant positive correlations were observed among the growth characteristics, suggesting that they are closely related. In contrast, no significant correlations were observed between the growth characteristics and SWV, indicating that they are independent. We conclude that mechanical properties are also important criteria for selecting plus trees in tree breeding programs. Principal component analysis revealed that seed provenances from Indonesia (Bangilan, Deling, and Randublatung) and India (Malabar and Central Province) have high scores of growth characteristics and SWV.  相似文献
